白芨胶载外源性rhEGF对伤口愈合影响的组织学研究 被引量:12

摘要 目的:探讨中药白芨胶(Bletillastriatagelatin,BSG)作为外源性重组人表皮生长因子(RecombinantHumanEpidermalgrowthfactor,rhEGF)的载体对兔背部伤口愈合的影响及其机制。方法:选取健康家兔40只,体重2.5~3kg,雌雄各半。距脊柱正中旁开2cm处做皮肤全层切口,面积约2cm×2cm,左右各3个。用已配置好的药液浸透双层纱布外敷伤口,每日换药一次。取材后进行伤口愈合时间、伤口愈合率、HE和Masson染色后的光镜组织学观察。结果:BSG+rhEGF的伤口愈合率和伤口愈合时间与SW组(Salinewater,SW)、BSG组和rhEGF组比较均有显著差异(P<0.05)。HE和Masson染色后光镜组织学观察发现BSG+rhEGF能促进炎症细胞渗出,肉芽组织、新生毛细血管和胶原纤维形成。结论:BSG载外源性rhEGF通过促进肉芽组织、新生毛细血管和胶原纤维的生成进而促进伤口愈合,其作用优于单用白芨和单用表皮生长因子。 Objective In order to find a new as well as effective way to heal the wound,the effects of exogenous Recombinant human epidermal growth factor(rhEGF) and Bletilla stroata gelatim(BSG)on the promotion of intention and the possible mechanism were investigated. Methods 40 healthy rabbits weighting approximately 2.5-3.0kg, male and female in half. After the backs was sterilized by general iodophors,the six full thickness skin incisions, measuring 2cmx2cm,were created on both sides of spinal column,apart from the spinal column and were 2cm.The wounds were covered with the double-layer gauzes which were soaked with reagents. Dressings were changed every day.Full-thickness wounds which were 2cm inside the wounds edge were taken respectively.Wound healing time,the wound healing rate and tissue sections were observed microscope after HE and Masson staining. Results BSG+rhEGF comparing with SW,BSG and RHEGF have significant difference on the wound healing rate and time (P〈0.05).The observation of HE and Masson staining showed that the BSG+rhEGF could promote cell's effusion,granulation tissue,newborn blood capillary and collagen fibers" formation. Conclusion The exogenous high molecular weight epidermal growth factor mixed with bletilia striata gelatin promotes the wound healing through stimulating the granulation tissue,new blood capillary and collagen fibers,which is surpasser than the BSG treated group and the rhEGF treated group.
