摘要
目的观察凋亡抑制蛋白survivin在脉络膜黑色素瘤中的表达及其与微血管密度(microvessel density,MVD)的关系,探讨survivin对脉络膜黑色素瘤血管生成的作用及意义。方法采用免疫组化SP法检测51例脉络膜黑色素瘤患者组织中survivin、CD34的表达,以CD34标记肿瘤微血管,并在显微镜下计数微血管密度。结果瘤旁组织中无survivin表达,survivin在脉络膜黑色素瘤表达阳性率为66.67%,其表达与脉络膜黑色素瘤病理分型、巩膜是否受侵相关(P<0.05),而与肿瘤位置、肿瘤最大基底直径无关;sur-vivin的表达与MVD呈正相关,survivin阳性表达组中MVD明显高于survivin阴性表达组(P<0.05)。结论survivin在脉络膜黑色素瘤的形成与发展中可能通过抑制凋亡和参与血管形成起着重要的作用,并且与患者预后关系密切。
Objective To explore survivin gene expression in choroidal melanoma and the relationship between its expression and mtcrovessel density (MVD) and probe into the effects and significance of survtvtn in the progress of chorotdal melanoma. Methods Using tmmunohtstochemtstry (SP method) to detect survtvtn and CD34 in 51 cases of chorotdal melanoma. The mtcrovessel was tmmunostatned with anti-human CD34 antibody,and the MVD was counted under the microscope. Results Survtvtn wasn' t detected in nearby tissue of tumor, but was detected in 34 cases (66.67%) of chorotdal melanoma. There was no correlation between survtvtn and the tumor location, as well as the largest tumor diameter. However significant correlation was found between survivin and cell type, as well as tumor invasion to the scleral emtssarta ( P 〈 0, 05 ). There was a positive correlation between survtvtn and MVD. The MVD were significantly higher in survtvtn-postttve chorotdal melanoma than in survtvtnnegative choroidal melanoma(P 〈0.05). Conclusion Survivin may contribute to the progression of choroidal melanoma by playing an important role in inhibiting apoptosis and enhancing angiogenesis. Survivin expression may be considered an important prognostic factor of choroidal melanoma.
