摘要
目的:探讨大肠癌组织Smad4mRNA水平与DNA倍体异常之间的关系。方法:用逆转录-聚合酶链反应(RT-PCR)技术测定大肠癌组织Smad4mRNA水平,利用流式细胞仪测定与DNA倍体异常相关的各项指数,并分析两者间的关系。结果:Smad4mRNA在大肠癌组织中的阳性表达率为62·31%,正常大肠黏膜组织阳性率为86·48%,两组间差异有统计学意义,χ2=6·80,P=0·009。大肠癌组织中Smad4mRNA与肿瘤大小有关,χ2=8·147,P=0·004;与组织分化程度有关,χ2=6·684,P=0·001;与浸润深度有关,χ2=3·907,P=0·048;与淋巴转移有关,χ2=8·051,P=0·005;与Duke’s分期有关,χ2=5·466,P=0·019。大肠黏膜组织恶变后,DNA异倍体细胞大量增加,PI和DI明显升高,处于S期及G2、M期的细胞比例明显升高,G0期细胞比例明显降低,Smad4mRNA的相对灰度值与DI无相关性,r=-0·216,P=0·080。与SPF、PI、G2M呈负相关性,r值分别为-0·305、-0·293和-0·320,P值分别为0·010、0·010和0·007;与G0/1呈正相关性,r=0·297,P=0·010。结论:大肠癌组织中细胞增殖异常活跃,可能与Smad4表达降低有关。
OBJECTIVE: To explore the correlation of Smad4 mRNA and cell prolifetous index in colonrectal carcinoma. METHODS.. The presence of Smad4 mRNA was measured by reverse transcriptase polymerase chain reaction (RT-PCR) from 69 colorectal carcinomas and 37 normal controls. The analysis of DNA ploidy heterogeneity was measured by flow cytometry (FCM). The correlation of Smad4 mRNA and DNA ploidy heterogeneity was statisticaly analysis. RESULTS: The positive percentage of Smad4 mRNA in colorectal carcinoma were lower than those of normal controls. The expression of Smad4 mRNA was related to histological differentiation, local, tumor size, Infiltration, lympy node metastasis and Duke's stage. Those cells which were provided with DNA ploidy heterogeneity in colorectal carcinoma were increased compared with normal colorectal tissues. The ratio of SPF, PI, Dland G2M was higher than those of normal, whereas G0/1 was lower. No correlation coμLd be estabilshed between Smad4mRNA and DI. Negative correlation coμLd be established between Smad4 mRNA and SPF, PI, GzM, r=-0.305, -0.293, -0. 320, P〈0.05. Positive correlation occurred in Smad4 mRNA and G0/1, r = 0. 297, P〈0. 05. CONCLUSIONS: The DNA ploidy abnormity was corerelated with descending of Smad4 mRNA in colorectal carcinoma.
