摘要
目的:优化提取荞麦茎叶中黄酮类化合物的方法。方法:通过与甲醇索氏提取法比较,确定使用绿色、环保、无毒害的乙醇作为提取试剂从荞麦茎叶中提取黄酮类化合物。通过对乙醇提取法进行优化,增加预处理步聚,并应用正交实验进行了条件优化。结果:最终确定了适于工业化放大的荞麦茎叶中黄酮类化合物的提取工艺:干燥粉碎样品中加入15倍原料重的30%乙醇溶液,30℃下浸泡6 h,然后加入15倍原料重的95%乙醇,在70℃下恒温浸提1 h,优化后的乙醇间接提取法较乙醇直接浸提法的效率提高12.5%。结论:确定并优化了荞麦茎叶中黄酮类化合物的绿色提取方法,为工业化应用奠定了基础。
Objective: To optimize the method of extracting flavone from mixture of stems and/or leaves of buckwheat. Methods: By comparing with methanol method, ethanol was used to extract flavone from mixtures of stems and/or leaves of buckwheat. Extra pre-treating step with low concentration of 30% ethanol was used to improve the extraction yields. Orthogonal experiment design was used to optimize the extracting method. Results: The extraction process was optimized as follows: mixing raw material 30% ethanol 15 times weight of with powders of stem and/or leaves of buckwheat in 30 ℃ for 6 hours, then adding raw material 95% ethanol 15 times weight of, putting the mixture in 70 ℃ for 1 hour,the yield was improved by 12.5%. Conclusion: One environmental friendly method of extracting flavone from mixtures of stems and/or leaves of buckwheat is founded and optimized for the basis of production of in industrial scale.
