骨髓间充质干细胞联合促红素治疗大鼠急性心肌梗死实验研究

摘要 目的:观察静脉移植骨髓间充质干细胞(MSC)联合促红素(EPO)治疗心肌梗死的可能性。方法:大鼠MSC培养后,制备成悬液备用。40只Lewis大鼠用异丙肾上腺素(ISO)制作急性心肌梗死模型,随机分为4组:A.MSC加EPO治疗组(n=10);B.MSC治疗组(n=10);C.EPO治疗组(n=10);D.心肌梗死对照组(n=10)。4周后,观察大鼠的心功能、血流动力学变化。结果:与对照组相比,MSC加EPO组、MSC组和EPO组的心功能明显改善,左室腔明显缩小(P<0.05),梗死面积减少;其中MSC加EPO组较MSC组和EPO组的心功能好(P<0.05)。结论:骨髓MSC联合EPO治疗大鼠急性心肌梗死取得了良好的效果,比单用效果好。 Objective: To investigate the effect of intravenous exogenous-transplantation mesenchymal stem cells(MSC) and erythropoietin(EPO) for the treatment of myocardial infarction. Methods: MSC were isolated from male Lewis rats ,cultured and made into cell suspension. After two subcutaneous injections of 85 mg isoproterenol (ISO) per kilogram of body weight to develop acute ischemic model ,40 male Lewis rats were divided randomly into four groups : A. acute myocardial infarction (AMI) +EPO + MSC intravenous transplantation group (n=10) ; B. AMI + MSC transplantation group (n=10) ; C. AMI + EPO group (n=10) ; D. AMI control group (n=10). After four weeks, echocardiography and hemodynamics of rat hearts were observed. Results: Compared with D, cardiac function was obviously improved in A,B and C (P〈0.05), left ventricular chamber and infarct size was obviously decreased(P〈0.05) ; Compared with B and C, A had better cardiac function(P〈0.05). Conclusion: intravenous exogenous-transplantation of MSC with EPO are effective in the treatment of myocardial infarction in rats.
