摘要
目的研究FY-10、依曲替酸对寻常型银屑病患者外周血中性粒细胞趋化功能的影响作用,探讨维A酸治疗银屑病的作用机制。方法采用密度梯度离心法分离寻常型银屑病患者外周血PMN,分别用FY-10、依曲替酸处理后用transwell趋化小室法检测PMN趋向IL-8以及MCP-3+IL-8的趋化功能改变。结果FY-10与依曲替酸从浓度10-9mol/L开始对银屑病病人外周血中性粒细胞趋化功能产生抑制作用,随剂量增加抑制作用先增强后减弱,FY-10在10-7mol/L浓度抑制作用最强,依曲替酸在10-7mol/L与10-6mol/L浓度之间作用最强。结论FY-10与依曲替酸可通过抑制PMN聚集影响银屑病病程。*
Objective To investigate effects of FY - 10 and Acitretin on peripheral blood neutrophils( polymorphonuclear leukocytes ; PMN) migration of patients with psoriasis vulgaris. Methods Isolated PMN from peripheral blood of psoriasis vulgaris, then observed the effect of FY - 10 and Acitretin on PMN and the chemotactic change of PMN against IL-8 and MCP-3 + IL- 8 with Transwell. Results Frome the dosage of 10^-9mol/L, FY - 10 and Acitretin inhibited chemotaxis of PMN for peripheral blood of psoriasis vulgaris. And according to the rising of the drug dosage the prohibit intensity increased first and then decreased. The former reached the most at 10^ -7 mol/L and the latter was between 10 ^-7 mol/L and 10^ -6 mol/L. Conclusion FY - 10 and Acitretin had effects on the process of psoriasis through prohibiting PMN from accumulating. MCP - 3 itself had no effect on chemoat- tracting of PMN, but MCP - 3 could synergize with IL - 8 to chemoattract PMN, and IFN - γ could further increase the chemotaxis.
