大鼠脑缺血再灌注损伤后高同型半胱氨酸血症和BrdU变化及通心络对其影响 被引量:1

摘要 目的:探讨大鼠脑缺血再灌注损伤(MCAO)后内源性神经干细胞的增殖分化情况,以及大鼠脑缺血再灌注损伤(MCAO)后血浆高同型半胱氨酸血症(Hcy)含量变化和通心络对其影响。方法:采用大鼠缺血再灌注损伤(MCAO)模型,应用放免方法观察缺血后3天、5天、14天以及30天Hcy含量变化,免疫组织化学方法观察缺血后缺血侧室管膜及室管膜下区(SVZ)、海马齿状回(DG)BrdU阳性细胞数的变化。结果:造模后通心络组BrdU阳性细胞荧光强度值明显高于脑缺血再灌注模型组,差异显著(P<0.05)。MCAO血浆Hcy浓度显著升高(P<0.05),通心络能降低Hcy浓度。结论:大鼠缺血再灌注损伤后可引起其缺血侧SVZ、DG区星形胶质细胞及神经细胞反应和增殖;而通心络可显著增加MCAO大鼠神经干细胞增殖分化能力。Hcy可能起着重要作用。 Objective : To investigate the change of homocysteins (HCY) and the differentiation of neural stem cells after middle cerebral artery occlusion in rats and the effects of treatment with Tongxinluo. Methods: The ischemia was induced by temporary middle cerebral artery occlusion (MCAO) . At 3, 5, 14 and 30 days after MCAO, the numbers of HCY was observed, In ependyma and subependymal region ( SVZ ), Hippocampus dentate gyms ( DG ), the numbers of BrdU was measured by immunohistochemistry. Results : Compared with the sham operation group, HCY were significantly increased in the MCAO in the day 5, 14 and 30 ( P 〈 0.05) . After the treatment with Tongxinluo, the number of HCY had significantly decreased in the MCAO. the number of BrdU was significantly increased as compared with the MCAO group ( P 〈 0.05 ) . Conclusions : This study demonstrates that the focal cerebral ischemia in rats results in a rapid response, from resident astrocytes and nerve cell of the SVZ and DG to the side of ischemia. Tongxinluo enhances the ability of the differentiation and proliferation of neural stem cell in MCAO ; HCY may play an important role in cerebral ischemic injury.
