摘要
目的:探讨红活麻有效部位LWB-6抗CIA关节炎的作用及机制。方法:在BALB/c小鼠体内建立II型胶原所致关节炎模型。采用隔日灌胃的给药方式观察LWB-6对关节炎发病和关节损伤的影响;并通过DC表型和DC T-bet基因mRNA的转录水平测定,探讨LWB-6抗CIA作用机制。结果:给药40d后发现,LWB-6呈剂量依赖性地降低CIA关节炎的发病率,与空白对照组相比有显著差异;足关节滑膜组织HE染色检查,未观察到炎性细胞浸润,关节间隙完好。20mg.L-1LWB-6对BALB/c小鼠骨髓来源DC T-bet mRNA转录水平有明显下调作用,FACS分析显示此部位对CD11c+DC表面分子MHC-II和CD86表达无明显影响。结论:LWB-6预防给药对CIA关节炎有明显的干预作用,此作用与负调控DC T-bet表达相关。
OBJECTIVE To investigate the preventive effect of the active component LWB-6 of Honghuoma on CIA model and the underlying mechanism. METHODS Collagen 11 induced arthritis was set up in BALB/c mice. LWB-6 was administered every other day, the incidence of arthritis and injury inside the joint was observed; the underlying mechanism was explored by detecting the phenotype of DC and mRNA level of DC T-bet. RESULTS After 40 days of treatment, the incidence of arthritis was significantly decreased by LWB-6 in a dose related manner, compared with that in the blank control group; No inflammatory cells infiltration was found in synovium by histological analysis, and the interspace of joint was intact. T-bet mRNA level in bone marrow derived DC was down-regulated by 20 mg. L- ^-1 LWB-6, and the phenotype expression of MHC-II and CD86 on CD11c+ DC was unaffected by LWB-6 by FACS analysis. CONCLUSION LWt3-6 shows obviously preventive effects on CIA model, and which is associated with the negative control of DC T-bet transcription.
