大黄提取片对高脂饮食诱导肥胖大鼠能量代谢的影响 被引量:6

Effect of Refined Rhubarb Tablet on Metabolic Parameters and Energy Metabolism in Skeletal Muscle of High Fat Diet Induced Rats

摘要 目的观察大黄提取片对高脂饮食诱导肥胖大鼠骨骼肌解偶联蛋白3(UCP3)及能量代谢的影响,以探讨大黄的减肥作用机制。方法给饮食诱导肥胖大鼠灌服低、中、高剂量大黄提取片(200、400、800mg·kg-·1d-1),测定大鼠肥胖指标,显微镜下观察脂肪细胞大小,应用Real-timePCR和Westernblot检测大鼠骨骼肌UCP3表达量,高效液相色谱法(HPLC)测定大鼠骨骼肌细胞能荷值(EC)。结果大黄提取片呈剂量依赖性的增强减肥作用,高剂量大黄提取片能明显减轻饮食诱导肥胖大鼠腹腔脂肪重量和体重(P<0.05),降低Lee’s指数(P<0.05),减小脂肪细胞最大直径与最小直径(P<0.05),明显增加骨骼肌UCP3表达(P<0.05),并降低骨骼肌细胞能荷水平(P<0.05)。结论大黄提取片可能通过增加肥胖大鼠骨骼肌中UCP3表达﹑促进细胞能量代谢而有效减肥。 Objective To explore the anti-obesity mechanism of Refined Rhubarh Tablet,we investigate the effects of Refined Rhubarb Tablet on uncoupling protein 3 (UCP3)expression and energy metabolism in skeletal muscle of high fat diet induced obese rat. Methods High fat diet induced obese rats were administrated with different dosages (200mg,400mg,800mg/kg·bw/d) of Refined Rhubarb Tablet for two months. Metabolic parameters,adipoeyte maximum diameter and minimum diameter were analyzed.UCP3mRNA and protein level in skeletal muscle were determined hy Real-time PCR and Western blot technique respectively. Analysis of skeletal muscle energy metabolism was performed by high performance liquid chromatography (HPLC). Results Refined Rhubarb Tablet showed dose-dependent anti-obesity effect. High dose of Refined Rhubarb Tablet treatment group presented with reduced abdominal fat and body weight(P〈 0.05),decreased Lee's index(P〈0.05). Adipocyte maximum diameter and minimum diameter were significantly less than those of control group (P〈0.05). High dose of Refined Rhubarb Tablet stimulated UCP3 Expression and decreased energy metabolism significantly (P〈 0.05). Conclusion Refined Rhubarb Tablet has significant anti-obesity effect via enhancement of UCP3 expression and energy metabolism.
