摘要
目的:建立高效液相色谱法测定滴眼液中更昔洛韦和利巴韦林的含量。方法:Diamonsil ^(TM)C_(18)色谱柱(250mm×4.6 mm,5μm,迪马公司);流动相:0.34%磷酸二氢钾溶液-甲醇(95:5),流速:1.0mL·min^(-1),检测波长:207nm,进样量:20μL。结果:更昔洛韦和利巴韦林分别在1.0~10.0μg·mL^(-1)和10.0~100.0μg·mL^(-1)范围内呈线性关系;相关系数分别为0.9999和1.0000。平均回收率分别为99.3%(RSD=0.69%)和99.6%(RSD=0.74%)(n=9)。结论:本法可同时测定更利滴眼液中更昔洛韦和利巴韦林的含量,方法简便、准确。
Objective:To establish an HPLC method for the assay of ganciclovir and ribavirin in Geng - Li eye drops. Method: The HPLC system consisted of Diamonsil^TM C18 ( 250 mm × 4. 6 mm, 5 μm) , and mixture of 0. 34% potassium dihydrogen phosphate solution - methanol(95:5 ) as mobile phase, at the flow rate of 1.0 mL · min ^- 1 and the detection wavelength of 207 nm and the injection volumn was 20 μL. Results:The assay displayed good lineary, the linear range of ganciclovir was 1.0 - 10. 0μg · mL^-1 ( r = 0. 9999 ) and that of ribavirin was 10. 0 - 100. 0 μg· mL^-1 ( r = 1. 0000 ) ( n = 6 ). The average recovery ( n = 9 ) was 99. 3% ( RSD = 0. 69% ) and 99.6% ( RSD = 0. 74% ). Conclusion :The method is simple, accurate and can be used for establishing quality standards and control of the eye drops.
