摘要
目的探讨糖尿病(DM)患者外周血淋巴细胞DNA损伤与维生素E(V itE)的关系。方法取32例DM患者和32例对照组的外周血,分离淋巴细胞。应用W estern b lot技术测定淋巴细胞热应激蛋白70(Hsp70)水平,用高效液相色谱仪测定血浆V itE的水平。结果糖尿病患者和对照组DNA损伤积分分别为33.9±14.5和14.9±8.1(每名计数25个细胞);糖尿病患者明显高于对照组(P<0.01)。2组的V itE水平分别为7.2±3.6和15.5±5.4(P<0.01)。将V itE水平与其DNA损伤程度进行相关分析,发现V itE水平与DNA损伤呈明显的负相关(r=-0.79,P<0.01)。结论糖尿病患者存在明显的DNA损伤,可能与高血糖对机体的损伤有一定的关系,但DNA损伤在糖尿病中的作用尚需进一步的研究。
Objective To explore the relationship of DNA damage in lymphocytes of the peripheral blood and the level of vitamin E in diabetic patients. Methods The peripheral bloods were got from 32 diabetic patients and 32 healthy persons, and lymphocytes were separated. Levels of HSP70 in lymphocytes were measured by Western blot, and levels of vitamin E were detected by high performance liquid chromatography (HPLC). Results DNA damage score was respectively 33.9 ±14.5 and 14.9 ± 8.1 in diabetic patients and healthy controls. It was higher in diabetic patients than in healthy control subjects(P 〈 0.01 ). The levels of vitamin E was respectively 7.2 ± 3.6 and 15.5 ± 5.4, it was a significant difference between two groups (P 〈 0.01 ). The levels of vitamin E had strongly negative correlation with DNA damage scores (r= -0.79,P〈0.01). Conclusion DNA damage is found in diabetes mellitus,it may be associated with the high blood glucose. But the value of DNA damage in diabetes mellitus needed further research.
