沙门氏菌属特异性单抗试剂对现场分离菌株的鉴定

摘要 对食品从业人员体检中获得的211株疑似沙门氏菌分离物经M肉汤培养后快速离心,以108菌/ml包板,60℃烘干,然后用沙门氏菌属特异性单克隆体进行间接ELISA测定。结果在对第一批170个菌株的检测中47株呈阳性反应、123株呈阴性反应。第二批43个分离株中31株呈阳性反应、12株呈阴性反应。两组结果与菌株来源单位的初步结果核对并进一步做系统生化鉴定,最终证明沙门氏菌属特异性单抗试剂对现场分离菌株的敏感性为98.7%,特异性为97.1%。 ore than 200 field isolates suspected as Salmonella spp when preliminary identification was conducted,were detected with genus specific MAbs to Salmonella spp.After being cultured in M-broth for 8-10 hours ,the organisms were centrifuged to a pellet and resuspended into 2 ml normal saline respectively.This was then heated in a boiling water bath for 10 min and adjusted to 10 8cell/ml with PH9.6 coating buffer.After the suspensions were coated into 24-well microtiter plate and dried at 60℃,an indirect ELISA was performed with the MAbs and HRP-labled anti-mouse IgM.At last,98.7% sensitivity and 97.1% specificity of the test were evaluated on the basis of final identification by biochemical procedure.The results showed that the MAbs would be very useful in the development of diagnostic kit for rapid detection of Salmonella spp.
