Agrobacterium tumefaciens strain A208SE harbouring a virus plasmid pTi37SE and binary vector PROA93 was used for the transformation of tobacco leafdiscs.Transgenic tobacco plaaes were obtained after optimizing different conditions of theleaf disc transformation including the establishment of high frequency plantletsdifferentiation from cultured tobacco leaf discs.The gus tissue-specific expression waschecked by histochemical method and the results showed that the gus gene regulated byCaMV35 s promoter expressed tissue-specific in transgenic tabaccos.
