摘要
目的探讨新生豚鼠海马神经干细胞在人工外淋巴液中分化为类毛细胞的最佳生长条件。方法将原代培养的新生豚鼠海马来源的神经干细胞进行体外培养(15%人工外淋巴液+DMEM/F12),分别接种于包被多聚赖氨酸和鼠尾胶原的盖玻片上使其分化,3周后对分化的细胞进行免疫荧光检测,分别计数产生表达毛细胞标记蛋白MysionⅦa阳性细胞的比例。结果神经干细胞在接种鼠尾胶原的盖玻片上生长,分化为MysionⅦa阳性细胞的比例为13.6%。但在接种多聚赖氨酸的盖玻片上生长,MysionⅦa阳性细胞的比例仅为1.9%。结论鼠尾胶原是适合新生豚鼠海马神经干细胞在人工外淋巴液中分化为表达毛细胞特异抗体细胞的生长基质。
Objective To observe the effects of different growth media on the differentiation of neural stem cells in vitro. Methods The neural stem cells were resuspended to the cover-glass which was coated with collagen type- Ⅰ or poly-lysine in 24-well culture plates. The growth and differentiation of NSCs were observed, then the cells were examined by hair cell marker with immunocytofluorescence 3 weeks later. Results A total of 13.6% of the cells grew in collagen type- Ⅰ media expressed immunoreactivity to Mysion Ⅶ a, but only 1.9% of the cells in the poly-lysine media expressed immunoreactivity to Mysion Ⅶ a. Conclusions Collagen type- Ⅰ is suitable for the differentiation of stem cells to haircell-like cells in vitro.
