脑缺血及高压氧治疗中血管内皮生长因子mRNA表达的研究 被引量:22

摘要 目的从基因水平研究高压氧(HBO)治疗对VEGFmRNA表达的影响。方法77只小鼠随机分为单纯缺血再灌注组、缺血再灌注+高压氧治疗组、假手术组。夹闭小鼠双侧颈总动脉,建立脑缺血再灌注动物模型。用RT- PCR方法检测各组织VEGF_(164)mRNA的表达。结果正常海马组织存在VEGF_(164)mRNA的表达。缺血再灌注3 h时海马组织VEGF_(164)mRNA的表达与对照组相比表达升高(P<0.05);48 h达高峰(P<0.05);72 h表达下降(P<0.05)。但在高压氧治疗后VEGF_(164)mRNA的表达均低于相同时间点的缺血再灌注组(P<0.05);72h表达升高(P<0.05)。结论HBO可通过增加VEGF_(164)mRNA的表达,诱导缺血半暗带及周边缺血脑组织血管生成,改善脑缺血缺氧。 Objective To study the effects of hyperbaric oxygen on the expression of VEGF mRNA in mice after cerebral ischemia-reperfusion. Methods The model mice were randomly divided into simple ischemia and reperfusion group, ischemia and reperfusion and hyperbaric oxygen treatment group, sham operation group. Cerebral ischemia- reperfusion models were established by clamping both common carotid arteries in mice. The expression of VEGF164 mRNA in various groups was determined with RT-PCR technique. Results The expression of VEGF164 mRNA was determined in normal hippocampi tissues. The expression of VEGF164 mRNA at 3hr in ischemia and reperfusion group increased more significantly than that of control group (P〈0.05) , the peak time was 48h (P〈0.05 vs control group), decreased significantly at 72hr (P〈0.05). The expression of VEGF164 mRNA in HBO group was lower than that of ischemia and reperfusion group (P〈 0.05) at the same time points. Conclusions Hyperbaric oxygen therapy can induce high expression of VEGF164 mRNA which induce angiogenesis in ischemic penumbra and the surrounding tissue of cerebral ischemia and improve cerebral ischemia and anoxia.
