应用串联质谱法快速测定尿液中氯胺酮及其代谢物的研究 被引量:2

摘要 目的:应用串联质谱(CI 离子源)法进行尿中氯胺酮及其代谢物的快速测定。方法:取尿液2~3mL,加入固体碳酸钠少量和丙酸酐80μL,超声5min,加入醋酸甲酯2mL,超声萃取10min 离心(4000r·min^(-1))3min,取上层有机相转入事先加入无水硫酸钠的样品瓶(4mL 容量)中,供 GC-MS/MS 测定。结果:本方法不仅适用于尿中氯胺酮原药的检验,同时满足尿中去甲氯胺酮、脱氢去甲氯胺酮的检验,回收率达90%以上,最低检出浓度为:5×10^(-8)g·L^(-1),GC-MS/MS 的最低检出限为0.05ng。结论:本方法可满足确证48h 以内是否吸食过氯胺酮。 Objective: The method of quick determination to ketamine and the metabolites in urine by GC - MS/MS (CI source) was introduced in the paper. Method:Few sodium bicarbonate and 80 mL of propyl acid anhydride were added to the urine sample(2 -3 mL). The mixture was ultrasonic extracted in 5 minutes,then,with 2 mL of methyl acetate was added,operated as the former step. After 10 minutes ultrasonic extration,the organic phase was added to a clean bottle of 4 mL capacity in which some sodium sulfate were added in advance. It was prepared for the determination by GC - MS/MS. Results:The method was not only used for the determination to ketamine in ufine, but also to the determination to demethylatingketamine and dethydro - demehylketamine. The average recoveries were above 90%. The detection limit of concentration was about 5 × 10^-8 g · L^-1 ,and the limit of detection of GC- MS/MS was 0. 05 ng. Conclusion.A person inhaled ketamine can be determinated by the method.
