高效液相色谱法测定血浆中万古霉素的浓度 被引量:12

摘要 目的:建立血浆样品中万古霉素的高效液相色谱测定法。方法:以甲硝唑为内标,采用 ZORBAX Eclipse XDB-C_(18)色谱柱(4.6 mm×150 mm,5μm),流动相为25 mmol·L^(-1)磷酸二氢钾溶液-乙腈(92.8:7.2),流速1.2 mL·min^(-1),检测波长230nm,柱温20℃。结果:万古霉素在0.5~100.0μg·mL^(-1)血浆浓度范围内线性关系良好(r=0.9996),最低检测浓度为0.25μg·mL^(-1)。日内、日间 RSD(n=5)分别为2.4%~4.4%,3.4%~5.5%;低、中、高3种浓度净化回收率分别为76.3%,81.5%,77.1%,方法回收率分别为99.3%,100.6%,101.1%。结论:本方法快速、简便,结果准确。 Objective:To establish an HPLC method to detemine vancomycin in human plasma. Method:Metronidazole was used as an interal standard. The chromatography was performed on a ZORBAX Eclipse XDB -C18(4. 6 mm × 150 mm ,5 μm)column with the mobile phase consiting of 25 mmol · L^-1 potassium dihydrogen phosphate- acetonitrile (92. 8: 7.2), the flow rate was 1.2 mL ·min^ - 1. UV detection was set at 230 nm and column temperature was 20 ℃. Results:The calibration curve was linear within the range 0. 5 - 100.0 μg· mL^-1( r =0. 9996). The limit of test was 0. 25 μg · mL^-1. Intra - day and inter - day RSD were 2. 4% -4. 4% and 3.4% -5.5% ( n =5 ) , The cleansed recoveries were 76. 3% , 81.5% , 77. 1% and method recoveries were 99.3% , 100.6% , 101.1% ,respectively. Conclusion:The method is rapid,simple and accurate.
