食品中虾青素的C_(18)-HPLC-PDA分离与鉴定 被引量:4

摘要 在装备了二极管阵列检测器(PDA)的高压液相色谱(HPLC)上,应用C18柱,冰凉花和虾中的主要酮基类胡萝卜素-虾青素可以被分离。分离条件为:色谱柱为DiamonsilTM(5μm,4.6mm×25cm,DikmaTechnology);流动相A为乙腈-水(90∶10,V/V),用磷酸调pH3.0;流动相B为乙酸乙酯;线性梯度洗脱:B在15min内由0%增加至70%;15~25min内B维持在70%;流速=1.0mL/min;检测波长=480nm;PDA波长范围=280~580nm;进样量=20mL。根据其与参比样品的色谱行为和光谱特征比较,样品中的虾青素可被鉴定。它在C18-HPLC-PDA上的定量亦成为可能。 By HPLC equipped with PDA, a major ketocarotenoid, astaxanthin, from the flower of adonis and shrimp was readily separated on a C18 column under the following conditions: Column: Diamonsil^TM (5μm, 4.6mm×25cm, Dikma Technology); mobile phase A=acetonitrile-water (90:10,V/V), pH=3.0 adjusted by phosphate; mobile phase B=ethyl acetate; linear gradient: B increased from 0 to 70% in 15 minutes and then kept constant in the following 10 minutes at the flow rate of 1.0mL/min, monitoring wavelength at 470nm; the wavelength range of PDA =280 -580nm; sample injection volume=20uL. According to the comparison of its chromatographic behavior and spectral characters with those of reference samples, astaxanthin from the samples was identified. Its quantification was therefore possible with C18-HPLC-PDA.
