吸烟对9-cis-RA诱导肺癌组织RAR_β转录影响的研究 被引量:1

摘要 [目的]观察9-顺维甲酸(9-cis-RA)处理后肺癌组织和对照肺组织中RARβ转录水平的影响,以及吸烟对9-cis-RA诱导肺癌组织RARβ转录的影响。[方法]应用组织块法培养肺癌组织和对照肺组织,采用RT-PCR法检测9-cis-RA处理24h前后RARβ的mRNA表达水平。[结果]无吸烟史患者肺癌组织RARβ的基础转录水平(0.5786±0.1316)显著低于对照肺组织(0.8015±0.0916)(P<0.05),有吸烟史患者肺癌组织RARβ的基础转录水平(0.6209±0.2132)和对照肺组织(0.6477±0.1326)无显著性差异(P>0.05)。经9-cis-RA处理24h后,无吸烟史患者肺癌组织RARβ的转录水平(0.7773±0.1160)显著升高(P<0.05),有吸烟史患者肺癌组织RARβ的转录水平(0.6612±0.1828)升高但差异无统计学意义(P>0.05)。无吸烟史和有吸烟史患者间肺癌组织RARβ的转录水平变化无显著性差异(P>0.05)。[结论]RARβ的表达下降可能与肺癌的发生发展有关。9-cis-RA诱导肺癌组织中RARβ的表达水平升高并且达到对照肺组织的表达水平,吸烟可能抵抗9-cis-RA对肿瘤细胞RARβ的诱导作用。 [Purpose]To investigate the impact of 9-cis-retinoic acid(9-cis-RA) on transcriptional changes of RARβ in lung cancer tissue and controlled lung tissue, and to evaluate the effects of smoking on 9-cis-retinoic acid induced RARβ transcription. [Methods] Cancer tissue and controlled lung tissue were cultured by tissue mass method. Incubated with 9-cis-RA for 24h, RARβ mRNA in tissue was detected by RT-PCR method. [Results]Transcription level of RARβ in lung cancer tissue from patients without smoking history (0.5786±0.1316) was decreased significantly than that in normal controls (0.8015±0.0916) (P〈0.05). Lung cancer tissue (0.6209±0.2132) and normal controls (0.6477±0.1326) in patients with smoking history showed no significant difference(P〉0.05). After treated with 9-cis-RA for 24h,RARβ transcriptional level (0.7773±0.1160) increased significantly in cancer tissue from patients without smoking history (P〈0.05), it also increased in tumor tissue with smoking history(0.6612±0.1828) but showed no significant difference (P〉0.05). The change of RARβ transcriptional level of lung cancer tissue showed no significant difference between patients with smoking and without smoking (P〉0.05). [Conclusion] Decreased expression of RARβ may associate with lung cancer. 9- cis-RA can significantly induce RARβ expression, and reach the transcriptional level of controlled lung tissue. Smoking might antagonize inductive effects of 9-cis-RA on RARβ expression in lung cancer tissue.
