摘要 目的 测定虫草培养基中腺苷和虫草素的含量。方法 采用HPLC法,色谱柱为Waters Nova—Pak C18柱(300mm×3.9mm,4μm);流动相为水-甲醇-醋酸(185:14:1);流速1.0ml·min^-1,检测波长260nm。结果 腺苷的线性范围为2.5.50.0μg·ml^-1(r=0.9999),回收率为104.5%,RSD=1.32%;虫草素的线性范围为6.5~130.0μg·ml^-1r=0.9999),回收率为106.1%,RSD=1.46%。结论 该方法灵敏、快速、准确,适用于虫草培养基中腺苷和虫草素的检测。 OBJECTIVE To establish an HPLC method for determining the contents of adenosine and cordycepin in Cordyceps militaris. METHODS The column was Waters Nova - Pak C18 ( 3.9 mm × 300 mm,4 μm), and mobile phase was water - methanol - acetic acid( 185 : 14 : 1 ) with flow rate of 1.0 ml. min-1 and detection wavelength of 260 nm. RESULTS The linear range was 2. 5 - 50.0 μg·ml^-1 ( r = 0.9999 ) for adenosine and 6.5 - 130.0 μg· ml^ -1 ( r = 0. 9999 ) for cordyecepin, respectively. The average recovery of adenosine was 104.5% with RSD of 1. 316% ; and that of cordycepin was 106.1% with RSD of 1.46%. CONCLUSION This method is rapid and accurate. It used to determine the adenosine and cordycepin in culture medium with satisfactory results.
