摘要
[目的]探讨BRMS1mRNA在乳腺癌中的表达及其与乳腺癌临床病理特征的关系。[方法]采用Trizol提取组织总RNA,将mRNA转录成cDNA,用实时荧光定量聚合酶链反应(RFQ-PCR)技术检测乳腺组织BRMS1mRNA表达,以BRMS1mRNA和GAPDHmRNA含量的比值表示BRMS1表达水平。[结果]乳腺癌组织中BRMS1mRNA的表达水平明显低于癌旁组织(P<0.01),也明显低于正常乳腺组织(P<0.01),BRMR1mRNA低表达与临床病理分期和淋巴结转移有关(P<0.05,P<0.01),而与患者年龄、癌肿大小、病理类型无关(P>0.05)。[结论]乳腺癌组织中BRMS1mRNA的低表达与乳腺癌的发展与转移有关,可能成为判断乳腺癌转移的一个标志物。
[Purpose] To investigate BRMS1 gene expression in breast cancer and its clinical significance. [Methods] Total RNA was extracted with Trizol. mRNA was transcribed reversely into cDNA. BRMS1 was detected with RFQ-PCR. BRMSI gene expression were presented as the ratios of BRMS1mRNA to GAPDHmRNA. [Results] BRMS1 expression in cancer tissue was significantly lower than that in para-cancerous tissue and in normal tissue (P〈0.01).BRMS1 low expression was related to clinico-pathologic stage and lymph node metastases (P〈0.05,P〈0.01,respectively), but not related to age,tumor size and pathologic type (P〉0.05). [Conclusion] BRMS1 low expression is related to the progression and metastasis of breast cancer, it might be served as a marker for breast cancer metastasis.
