E2F-1在胃癌组织中突变与表达的研究 被引量:1

摘要 目的研究E2F-1在胃癌组织中的基因突变、表达情况。方法实验分为3组:胃癌组、癌旁正常黏膜组和正常对照组。采用聚合酶链式反应-单链构像多态性分析法(PCR-SSCP)对3组的E2F-1基因突变情况进行分析,采用免疫组织化学法检测三组中E2F-1的表达情况。结果PCR扩增产物电泳显示胃癌组、癌旁正常黏膜组和正常对照组均未发现E2F-1基因突变或缺失,SSCP分析3组所有外显子均未出现异常条带;胃癌组中E2F-1的阳性表达率为72.5%(58/80),癌旁正常黏膜组和正常对照组的E2F-1阳性表达率分别为30.0%(24/80)和22.5%(9/40);胃癌组E2F-1表达率明显较其他两组高,3组间差异有显著性(P<0.05)。结论E2F-1在胃癌的发生发展过程中是一个非常稳定、保守的基因,突变发生率极低;E2F-1在胃癌组织中呈高表达。
