摘要
目的:探讨雄激素对扩张型心肌病慢性心力衰竭雄性大鼠心肌细胞凋亡的影响。方法:将74只雄性SD大鼠分为:正常对照组(A组)、心力衰竭组(B组)、睾丸切除加心力衰竭组(C组)、睾丸切除加心力衰竭加睾酮替代治疗组(D组)。通过超声心动图观察大鼠左室收缩功能的变化,应用放射免疫法检测大鼠血浆神经内分泌激素(睾酮)和肿瘤坏死因子-α水平,用凋亡原位检测方法(TUNEL法)检测心肌组织凋亡细胞,计算凋亡指数。结果:与B、C组比较,D组肿瘤坏死因子-α水平明显降低,心肌细胞凋亡指数减少,心功能也明显改善。结论:雄激素能抑制扩张型心肌病雄性大鼠心肌细胞的凋亡,从而保护了心功能,其作用的一个重要机制即是雄激素能拮抗肿瘤坏死因子-α的产生。
Objective:To investigate the effects of androgen on apoptosis of myocardial cells of male rats with chronic heart failure(HF). Method:Seventy-four SD male rats were divided into 4 groups: control(A group), HF (B group), orchiectomy(ORX) +HF(C group), and ORX+HF+Testosterone(T) (D group). The left ventricular function was examined by echocardiography, the level of testosterone and cytokine (TNF-α) were measured by radioimmunoassay, and the apoptosis was evaluated histochemically by in situ end-labeled DNA(TUNEL method). Result:Compared to C group, there were better cardiac performance and greatly decreased apoptosis index of myocardial cells in D group. Meanwhile the plasma levels of TNF-α in D group was significantly decreased compared to that of C group. Cohclusion:In male rats with dilated cardiomyopathy, testosterone exerts a suppressive influence on apoptosis of myocardial cells mostly due to its inhibition on TNF-α.
