摘要
以南方红豆杉为材料,建立了其愈伤组织诱导与保持的技术体系,主要为:(1)南方红豆杉侧芽是诱导愈伤组织的最佳外植体;(2)南方红豆杉愈伤组织诱导的最佳培养基为:B5+2.0 mg.L-12,4-D+1.0 mg.L-1NAA+0.2 mg.L-1IAA;(3)南方红豆杉愈伤组织增殖的培养基为:B5+4.0 mg.L-1NAA+0.5 mg.L-12,4-D+0.2 mg.L-1GA+500 mg.L-1AC+2000 mg.L-1LH.并在培养基中同时添加AC、LH和GA33种抗褐物质,取得较好的抗褐化效果.
The callus induction and maintenance system of Taxus chinensis var. mairei was established in the experiment, the main results were as follows: (1). lateral buds were the best explants for the callus induction; (2) B5 plus 2.0 mg · L^-l 2,4-D, 1.0 mg · L^-1 NAA and 1.0 mg · L-1 IAA was the best medium for callus induction; (3) B'5 containing 4.0 mg· L-1 NAA, 0.5 mg· L^-1 2,4-D, 0.2 mg · L^-1 GA3 , 500 mg· L^-l AC and 2000 mg·L^-l LH was the best medium for callus growth. The brownness of callus could be avoided or decreased to a certain degree when AC, LH and GA3 were added to the mediums.
