摘要
目的应用鼻内滴入卵白蛋白(OVA)法建立大鼠免疫耐受模型。方法OVA免疫耐受组连续3d鼻内滴入卵白蛋白,继之致敏和激发;哮喘组直接致敏和激发。于末次激发24h后收集支气管肺泡灌洗液(BALF),计数细胞总数并分类;应用ELISA检测上清中IL-4、IL-5和IFN-γ的含量;采集血清并用ELISA法检测OVA特异性IgE含量;应用HE染色行肺组织病理学检查。结果鼻内滴入OVA后大鼠BALF中细胞总数及嗜酸性粒细胞(Eos)、淋巴细胞和中性粒细胞百分率明显减少,血清中OVA特异性IgE含量及BALF上清中IL-4和IL-5的含量明显降低,IFN-γ的含量明显升高,肺组织中细支气管周围炎症细胞浸润和黏液的过度分泌明显减轻。结论鼻内滴入OVA可以成功建立大鼠免疫耐受模型。
[Objective] To establish the rat model of immuine tolerance through intranasal treatment with oval bumin. [Methods] Rats in immuine tolerance group received OVA intranasally on each of 3 consecutive days, followed by sensitization and challenge. Asthma group received sensitization and challenge only, 24 h after the last challenge, bronchoalveolar lavage fluid (BALF) was collected, and cytology study was conducted. The IL-4, IL-5 and IFN-γ BALF levels and the OVA-specific serum IgE level were determined by ELISA. The lung tissue was obtained and histologic analysis was done through HE. [Results] Total cells number, the percentage of eosinophils (Eos), lymphocytes and neutrophils in BALF, the IL-4 and IL-5.BALF levels and the OVA-specific serum IgE level of immuine tolerance group were lower than that of asthma group, while its BALF IFN-γ level was higher than that of asthma group. Compared with asthma group, peribronehiole inflammatory cells infiltration and muein production of immuine tolerance group were alleviated. [Conelision] Intranasal administration with ovalbumin can establish rats model of immuine tolerance successfully.
