转化生长因子β1基因多态性对煤工尘肺遗传易感性的影响 被引量:6

THE EFFECT OF THE TGF-β1 GENE POLYMORPHISM ON GENETIC SUSCEPCEPTIBILITY OF COAL WORKER's PNEUMOCONIOSIS

摘要 目的:探讨转化生长因子β1(TGF-β1)基因-509位点多态性对煤工尘肺遗传易感性的影响。方法:以110例汉族煤工尘肺患者和110例汉族煤尘接触者为研究对象,进行问卷调查,采集其外周静脉血提取DNA并应用PCR-RFLP技术检测其TGF-β1基因-509位点多态性。结果:煤工尘肺患者中TGF-β1基因-509位点为C/T及T/T基因型的占81·8%,C/C基因型占18·2%;而煤尘接触者中则分别为53·6%和46·4%,两组之间差异有统计学意义(P<0·05)。调整接尘工龄后,C/T及T/T基因型罹患煤工尘肺危险度是C/C基因型的3·308倍(P<0·05)。logistic回归分析结果显示TGF-β1基因-509位点基因型、接尘工龄、吸烟3个因素均对罹患煤工尘肺有影响。结论:TGF-β1基因-509位点C/T及T/T基因型可能比C/C基因型者易于罹患煤工尘肺,接尘工龄、吸烟亦是煤工尘肺重要的影响因素。 Objective: To approach the effect of transforming growth factorⅠ (TGF - β1) gene polymorphisms in site of - 509 on genetic susceptibility of coal worker' s pneumoconiosis (CWP) .Methods: 110 cases of coal worker' s pneumoconiosis and 110 coal mine workers (controls) were selected and investigated by questionaire . DNA was extracted from 3 ml peripheral vein blood that was drawn from every subject.Polymerse chain reaction-restriction fragment length polymorphisms (PCR-RFLP) techniques was usecl, TGF-β1 gene polymorphisms in site of -509 were analyzed.Results: There was a significant difference between CWP workers and controls in the distributive frequencies of the TGF- β1 genotypes in site of - 509 (P〈0.05) . In the cases of CWP, the distributive frequency of the TGF- β1 gene with genotypes C/T and T/T was 81.8 %, the distributive frequency of the genotype C/C was 18.2%. In the coal mine workers (controls), the two distributive frequencies were 53.6% and 46.4% .When the length of service of the subjects was controled , relativing to the genotype C/C, the ORadj of the genotypes C/T and T/T to CWP was 3.308 (P〈0.05) .Conclsions: The TGF- β1 gene polymorphisms in site of - 509 might play an immportant role in CWP . TGF - β1 gene in site of - 509 with genotypes C/T and T/T would promote the genetic susceptibility of CWP.
