在RAPD扩增的基础上,获得了一个香菇(Lentinulaedodes)135菌株的SCAR(SequenceCharacterizedAmplifiedRegion)标记。为了验证这一标记的准确性,在163个香菇菌株上进行了SCAR扩增,鉴定出9个都含有135SCAR标记的菌株。为进一步了解这些菌株的遗传背景关系,对包括Q135和F135在内的11个135系列菌株进行了拮抗实验和AFLP分析,结果显示,大多数菌株间无拮抗反应,少数有弱拮抗反应;AFLP分析得知,这一系列菌株间有着很高的相似性,平均相似系数为0.9271,它们都来源于福建三明真菌所的135菌株,各地引进后出现了一些变异,但由于引进年限不长,还没有形成明显的生理小种。因此,利用RAPD-SCAR技术获得的135SCAR标记用于菌株鉴定是可行和可靠的。
The accuracy of a specific Sequence Characterized Amplified Region (SCAR) marker for Lentinula edodes ‘135’ strains, obtained using primers based on RAPD methodology, was confirmed by using the marker to correctly identify nine ‘135’ strains from a test sample of 163 L. edodes strains. Antagonism tests and AFLP amplification were used to further analyze these nine ‘135’ strains, together with strains Qingyuan 135 (Q135) and Fujian 135 (F135), in order to better understand their genetic background. A total of seventeen antagonistic reactions were recorded although the degree of the antagonism observed was generally weak. AFLP analysis revealed a high similarity between the eleven ‘135' strains, and the average similarity coefficient was 0. 9271. Our data demonstrate the feasibility and reliability of the specific SCAR marker for distinguishing ‘135’ strains from other L. edodes strains.
