温郁金对人胃癌裸鼠原位移植瘤生长转移的抑制作用及对COX-2和VEGF表达的影响 被引量:19

摘要 目的:通过建立人胃癌裸鼠原位移植瘤模型,研究移植瘤生长及其转移的抑制作用。方法:BALB/c裸小鼠12只,以荷SGC-7901人胃癌胞株裸鼠皮下移植瘤为材料,通过手术将癌组织转移到裸小鼠胃壁建立胃癌原位移植模型;造模后,12只荷瘤裸鼠随机分为两组(对照组、治疗组),每组6只;原位移植24h后开始灌胃,治疗组给予温郁金水蒸气蒸馏液(2.5g/mL),0.4mL灌胃,每日1次,对照组予等量生理盐水。当动物出现明显消瘦、弓背、神萎等衰竭征象(约9周)时处死动物,观察和比较两组肿瘤抑制率和肿瘤转移情况。用免疫组化法检测两组移植瘤瘤体中COX-2和VEGF的表达。结果:所有荷瘤鼠胃壁均有肿瘤生长,荷瘤率100%,治疗组与对照组瘤重分别为(0.4189±0.1086)g、(0.7296±0.210)g,P<0.01,抑瘤率为42%;模型组腹膜转移6/6、肝脏转移5/6、腹水1/6,治疗组腹膜转移2/6、肝脏转移1/6、腹水0/6,其中腹膜及肝脏转移情况差异具有统计学意义(P<0.05)。温郁金治疗组瘤体中COX-2和VEGF的表达明显下调。结论:温郁金具有抑制人胃癌裸鼠原位移植瘤生长和转移的作用,其作用机制可能与下调COX-2和VEGF的表达有关。 Objective :The aim is To investigate the effects of common turmeric (wenyujin) on the nude mouse model of human gastric carcinoma cells using orthotopic transplantation and its possible mechanism. Methods:Firstly we established nude mouse orthotopie implant/metastatic model of human gastric cancer. Human gastric adenocareinoma cell line ( SGC - 7901 ) was inoculated subcutaneously into nude mice to develop implant tumor. Intact tumor tissue was then collected and implanted orthotopieally into the stomach of nude mouse. On the second day The twelve experimental, mice were derided into two groups at random,6 in expertmental group and 6 in control group, the cxperimental group was administrated with a dose of 0.4mL. vsporized distilled common turmeric (the concentration was 2.5g/mL)once a day, while the control group was administrated with the same dose of saline meanwhile. When they became sick about nine weeks later , they were killed. The inhibitary rates of tumour growth were detected , and local invasiveness, the rates of metastasis in liver and peritonetma were compared between the two groups. By immunohistochemical methed, the expression of vascular endothelial growth factor(VBGF) and eyelooxygenase -2 (cox -2) in gastric carcinoma were determined. Results :The rate of successfully orthotopic transplantation was 100%. The weight of the tumors in the experimental in the experimental group was less than that of the control group significantly( 0.4189 ± 0. 1086 )g vs (0. 7296 ± 0.2104) g, P 〈 0.01, The inhibitary rates of the experimental group was 42%. In the face of tumor metastasis, the rates of metastasis in peritoneum,liver and ascites of the control group were 6/6,5/6,1/6, those in experimental group were 2/6,1/6,0/6, respectively. Compared with the conhrol group, the rates of metastasis of peritoneuma, liver in the experimental group were signficantly lower( P 〈 0.05 ). meanwhile we found that the positive rate of COX -2 and VEGF in the experimental groups were obviously lower than these in control group( P 〈 0. 05 ). Conclusions : Common turmeric has a strong inhibitory effect on both growth and metastasis of gastric cancer, and the mechanism of that may be related with down - regulation of COX - 2 and VEGF.
