摘要
以马铃薯栽培品种甘农薯1号的试管苗为材料,进行了茎尖超低温保存技术的研究,结果表明,预培养3~4d,用玻璃化溶液PVS2室温下处理20~30min,直接投入液氮中冷冻保存,然后接种在MS添加2.50mg/LKT、1.25mg/LIAA和1.25mg/L6-BA的培养基上,经过恢复培养,茎尖成活率可达42.8%。
Shoot tips of plantlet in vitro of potato cultivar Gannongshu 1 as donors, cryopreservation of shoot tips was studied in the experiment. The excised shoot-tips, were precultured 3-4 d and treated by vitrification solution PVS2 for 20- 30 rain under room temperature, plunged into fluid nitrogen and stored for a certain time then transferred onto MS medium supplemented with 2. 5 mg/L KT, 1.25 mg/L IAA and 1.25 mg/L 6-BA, survival rate of shoot tips of potato could reached 42.8%.
