甘薯愈伤组织对干旱胁迫的生理反应研究 被引量:8

摘要 以“芦选一号”甘薯盆栽苗叶片为外植体进行组织培养,建立甘薯愈伤组织诱导和扩繁体系,并以从叶片诱导产生的均匀一致愈伤组织为材料,研究干旱胁迫对细胞膜透性、可溶性糖、脯氨酸含量、SOD、POD、CAT活性的影响,从而在细胞水平上探讨甘薯抵御干旱胁迫的生理机制,主要结果如下:干旱胁迫下,甘薯愈伤组织的膜透性显著增大;中度和重度胁迫下,SOD活性升高;不同胁迫程度处理下,POD活性普遍下降;中度和重度胁迫下,CAT活性普遍升高;不同胁迫程度处理下脯氨酸和可溶性糖含量普遍升高. Leaves from sweet potato Lu-xuan 1 grown in pot are used as explants in tissue culture, to establish the induction and proliferation system of sweet potato callus. Then, the homogeneous callus from leaves is cultured under drought stress, and then their effects. On Plasma membrane permeability contents of soluble sugar and pro, SOD PODandCAT activities were determined, so as to understand the physiological mechanisim of sweet potato callus resistance to drought stress at cell level. The results are presented as follows: The permeability of cytoplasm membrane of sweet potato callus rises prominently under drought stress; SOD activity increases under moderate and severe stress; POD activity decreases in most treatments under different degree stresses; CAT activity increases in most treatments under moderate and severe stress; and contents of Pro and soluble sugar increases in most conditions under different degree of stresses.
