软脂酸对美吡哒和高糖刺激大鼠胰岛β细胞功能抑制作用的研究

摘要 目的观察软脂酸对美吡哒和高糖刺激的大鼠胰岛细胞β细胞功能的影响。方法以0、0·25、0·5、1·0mmol/L软脂酸原代培养SD大鼠胰岛细胞,在24h、72h、120h测定上清胰岛素、葡萄糖浓度及细胞内胰岛素。结果在不同培养时间不含软脂酸组上清中胰岛素、葡萄糖及细胞内胰岛素含量差别无统计学意义(P>0·05);而含软脂酸组随软脂酸浓度增加和培养时间延长,胰岛素分泌量、葡萄糖利用量和细胞内胰岛素含量逐渐减少(P<0·05)。结论在体外软脂酸抑制了美吡哒和高糖刺激的大鼠胰岛β细胞功能,这种抑制作用与胰岛细胞本身胰岛素抵抗相关。 Objective To observe the effect of different concentrations of palmific acid on the function of rat pancreatic β-cells in different periods stimulated by glipizide and glucose in vitro. Methods Different concentrations of palmitic acid were contained in supematant of medium and insulin contents in islet cell were measured in different periods of cultivation. Results In groups Islet cells were cultured without palmitic acid,there were no statistically significant differences among insulin secretions,utilizations of glucose and insulin contents in islet cell. There were statistically significant differences in the other groups. Conclusions Palmitic acid inhibits the function of mt pancreatic β-cells stimulated by glipizide and high concentration of glucose in vitro. The inhibition is closely related with insuin resistance of islet cells.
