瑞香狼毒和狼毒素对体外培养小鼠白血病P_(388)细胞生长的影响 被引量:27

Effects of Chinese Chellera (Stellera chamaejasme L.) and Chamaejasinine on Proliferation of Mouse Leukemia P388 Cells in Vitro

摘要 瑞香狼毒水提物(ScL)对体外培养的小鼠白血病P_(388)细胞生长、集落形成和MTT甲化合物生成有显著抑制作用,并和剂量呈正相关。0.25~169·L^(-1)处理细胞24h,细胞生长抑制率为2.8~88.4%,MTT甲化合物生成抑制率为3.1~73.0%,处理细胞12h,P_(388)细胞软琼脂集落形成率减少0.9~70.9%。从瑞香狼毒中提出的狼毒素(Cha)对体外培养的小鼠白血病P_(388)细胞生长的抑制作用较ScL弱,0.25~50mg·L^(-1)处理细胞24h,细胞生长抑制率为1.6~32.8%;提示Cha不是ScL的主要抗肿瘤活性成分。 The effects of Chinese Chellera ( Stellera chamaejasme L., Scl. ) and its compound chamaejasmine (Cha) on proliferation, clonal formation and MTT formazan formation of P338 cells in vitro were studied. It was found thet ScL inhibited the proliferation of P338 cells, and the inhibition rate had a positive interrelationship with the concentra- tion and exposure time. At the concentration of 0.25- 16 g·ml-1 , inhibition rate was 2.8-88.4% . After exposure of the cells to ScL 2g·ml-1 for 6, 12, 24, 48h, the inhibition rates were 7.4, 19.9, 39.1, 59.1% respectively. ScL inhibited also markedly the MTT formazan formation and clonal formation of P388 cells. The inhibitory effects of Cha on the proliferation, clonal formation and MTT fromazan formation were weaker than that of ScL. After exposure of the cells to Cha 0.25-50mg·ml-1 for 24h, the inhibition rate was 1.6-32.8% . These results showed that the antitumor activity of ScL was produced on the basis of its cytotoxin effects. Chamaejasinine is not the main antitumor agent of ScL.
