不同药物对动脉粥样硬化兔体内两种受体表达的影响

Effect of different medicament on the receptor expression in the atherosclerotic rabbits

摘要 为了给药物的筛选和利用提供基础数据,用手术法切除新西兰雌兔双侧卵巢,用高胆固醇饮食法制备动脉粥样硬化模型。利用实时荧光定量RT-PCR方法,对动脉粥样硬化兔心脏和肝脏内ER mRNA和LDLRmRNA的表达进行了分析。研究表明,ERα在模型组(C)、假手术组(SC)、正常饲料组(N)、利维爱组(TIB)、倍美力组(PRE)和新伐他汀组(SIM)心脏内的相对表达量分别为0.29、0.53、0.46、0.85、0.48、0.30,在肝脏内的相对表达量分别为0.32、0.51、0.49、0.68、0.50、0.30;低密度脂蛋白受体(LDLR)在模型组(C)、假手术组(SC)、正常饲料组(N)、利维爱组(TIB)、倍美力组(PRE)和新伐他汀组(SIM)心脏内的相对表达量分别为0.22、0.24、0.33、0.27、0.31、0.23,在肝脏内的表达量分别为0.86、0.93、1.52、1.27、1.19、0.88。结果显示,利维爱通过增加心脏和肝脏内雌激素受体、低密度脂蛋白受体的表达发挥抗动脉粥样硬化的作用。新伐他汀可以有效的防治动脉粥样硬化的发展,对心脏和肝脏内的2种受体的表达没有影响。倍美力防治AS的效果不明显,可以促进ERα和LDLR在心脏和肝脏内的表达。 Atherosclerotic animal model with oophorectomized rabbits and high cholesterol feedstuff was made. With real- time fluorescence quantitive RT- PCR, we studied the expression of estrogen receptor a and low density lipoprotein receptor mRNA in heart and liver from atherosclerotic rabbits. The results showed: the relative expression of ERa in heart from model group (C), sham- operation group (SC), standard chow group (N), tibolone group (TIB), premarine group (PRE), Simvastatin group (SIM)was 0.29, 0.53, 0.46, 0.85, 0.48, 0.30 respectively,and was 0.32, 0.51,0.49,0.68,0.50,0.30 respectively in liver; the relative expression of LDLR in heart from group C, group SC, group N, group TIB, group PRE, group SIM was 0.22,0.24,0.33,0.27, 0.31,0.23 respectively, and was 0.86,0.93,1.52,1.27,1.19, 0. 88 respectively in liver, Livial, has the function of anti - atherosclerosis by stimulating the expression of ERα and LDLR mRNA. Simvastatin can reduce the progression of AS, whereas has no effect on the two receptors. Premarine increased the level of ERα and LDLR mRNA in heart and liver, but has no significant effect on AS plaque.
