介绍一种大量纯化钙调素的方法

A Large Scale Purification Method of Calmodulin

本文介绍了一种适于大量制备钙调素（ＣａＭ）的简易实用方法。我们用该方法从２．５ｋｇ猪脑组织中一次纯化了１５０ｍｇＣａＭ。经鉴定，所提ＣａＭ的纯度在ＳＤＳＰＡＧＥ中呈单一区带，分子量为１９ｋＤ（在１ｍｍｏｌ／ＬＥＧＴＡ的条件下），等电点为４．３８，并对ＰＤＥ有明显的激活作用。该作用可被特异性抑制剂ＴＦＰ抑制。

simple and a large amount purification method of calmtalulin(CaM)from porcine brain wasperformed by modified hydrophobic affinity chromatography. With this method,150mg CaM from 2.5kg porcine brain was prepared.The purity,molecu-lar weight,isoelectric point,and activating effect of the product on PDE were studied.This method has following advantages: The whole precedure was more simple and efficientthat 60 mg CaM could be obtained from 500g porcine brain,By this method,the amount of caMwas yielded more higher than that of others.
