摘要
目的探讨茶多酚抑制口腔上皮癌细胞增殖的作用及其相关机理。方法建立茶多酚抑制肿瘤细胞生长的体外模型,采用MTT法和流式细胞仪技术,体外观察茶多酚对口腔上皮癌细胞增殖抑制及相关蛋白表达的影响。结果不同浓度的茶多酚(50、100、200、400μg/m1)对口腔上皮癌细胞均有抑制作用,呈剂量依赖关系,抑制率分别为44.5%,58.2%,74.5%和79.7%。在细胞增殖受到明显抑制时,细胞被阻滞于G0/G1期,不能进入S期及G2期。与对照组比较,随着茶多酚浓度的增高,细胞表面CD151表达水平逐渐增高,CD82表达水平逐渐降低。结论茶多酚可抑制口腔上皮癌细胞增殖,其作用机制与改变细胞表面CD151和CD82蛋白表达水平有关。
Objective To explore the mechanism of inhibitory effect of tea polyphenols (TPP) on proliferation of oral epithelial carcinoma KB cells. Methods Set up anti-tumor model by culturing KB cells with different concentrations of TPP. MTT assay was performed to determine the inhibitory rate of KB cells. Flow cytometry analysis was applied to measure the expression of PETA-3/CD151 and KAI-1/CD82 on the surface of KB cells. Results TPP inhibited proliferation of KB cells in a dose-dependent manner. Under the dosage of 50ug/ml, 100ng/ml, 200ug/ml and 400ug/ml TPP, the inhibitory rates were 44.5%, 58.2%, 74.5% and 70.7% respectively. The inhibited KB cells were blocked in G0/G1 phase, and could not transfer to S or G2/M phase. Compared with the control group, expression of PETA-3/CD151 and KAI-1/CD82 gradually decreased with the increase of TPP concentration, respectively. Conclusions TPP can inhibit the proliferation of KB cells and the mechanism is related to the changes of PETA-3/CD151 and KAI-1/CD82 expression.
