龙胆中龙胆总苷的含量测定

Contents Mensuration of Totalglycosides of Radix Gentianae

目的：建立一项对龙胆中龙胆总苷含量测定的分析方法。方法：采用紫外-可见分光光度法（uv-2450），以D101大孔吸附树脂纯化样品（1．5cm×10cm），1％变色酸为显色剂，酸度控制在65％，沸水加热，显色时间为25min，显色最大吸收波长为576nm。结果：龙胆苦苷在0．0410～0．1093mg·mL^-1质量浓度内线性关系良好（r=0．99993），方法回收率为98．8249％，RSD为0．46％．结论：该方法可用于质量控制，为龙胆中龙胆总苷含量测定提供了一项可行的方法，进而也为龙胆中间提取物质量控制提供了一项总苷含量测定的方法。

Objective： To establish a determined method for the contents mensuration of totalglycosides of Radix Gentianae. Methods： Gentianae totalglycosides was purified with D101 macroreticular resin （1.5cm × 10cm） and was determined by Chromotropic Acid （ 1% ） - suffuric acid method and detected at 576nm by UV. Results ： The calibration curve was linear （r = 0.99993） in the range of 0.0410 - 0. 1093mg·mL^- 1 for gentiopicroside, the average recovery of the method was 98. 8249%, with RSD 0.46% （ n = 5）. Conclusions： This method can be used for the quality study and contents mensuration of Radix Gentiance, this method can be also applied with contents determination of the middle - extracts of Radix Gentianae.
