摘要
目的:探讨肿瘤细胞诱导分化剂六亚甲基二乙酰胺(HMBA)和N-乙酰-1,6-二胺基己烷(NADAH)与人红白血病K562细胞DNA的相互作用和诱导肿瘤细胞分化的机理.方法:通过两步合成,将NADAH偶联到对甲基苯磺酰氯活化的异丙基甘油硅胶上,制备高效液相色谱柱,研究人红白血病K562细胞DNA在NADAH柱上的行为及NADAH及HMBA与人红白血病K562细胞DNA的相互作用.结果:以2mol/L(NH4)2SO4pH7.4的磷酸缓冲液做降盐梯度洗脱时,K562细胞DNA大部分很快被洗脱流出,另有一小部分DNA在(NH4)2SO4浓度降至最低时才流出,NADAH柱对这部分DNA的保留能力极强,即有很强的疏水作用;不同的DNA与NADAH具有不同的疏水亲和力.NADAH柱对DNA的离子效应很弱.结论:HMBA和NADAH在诱导肿瘤细胞的分化过程还对DNA有疏水作用.
Objective: To study the interaction between N acetyl 1,6 diaminohexane (NADAH) and DNA of the K562 cells and the mechanism of NADAH and hexamethylene bisacetamide (HMBA) inducing tumor cells differentiation. Methods: NADAH was immobilized onto the diol silica by two step synthesis. Then HPLC NADAH column was prepared. The interaction between NADAH and DNA of the K562 cells was analyzed on NADAH column. Results: Of K562 cells DNA, some have very strong hydrophobic interaction since these were not eluted out until the ion intensity was decreased below that of the normal physiology, but no notable ion exchange was observed. Conclusion: These results suggested that HMBA and NADAH should exist the hydrophobic interaction with some DNA in the process of their inducing tumor cells differentiation.
