摘要
为探讨癌基因扩增在乳腺癌中的临床意义，应用差异ＰＣＲ技术检测Ｃ－ｅｒｂＢ２基因在７５例人乳腺癌中扩增情况。结果：３４．７％（２６／７５）的乳腺癌存在该基因扩增，且以ＥＲ阴性或腋淋巴结转移＞３枚者明显，而与其它因素关系不大。通过Ｓｏｕｔｈｅｒｎ印迹杂交对原样本再次检测，结果与上述基本一致，并发现一例基因重排。表明应用差异ＰＣＲ技术检测Ｃ－ｅｒｂＢ２扩增具有快速、简单、可靠、无放射性污染，有助于乳腺癌预后及转移潜力之判断，临床应用价值较大。
Inordertoaddresstheclinicalimplicationofoncogeneamplificationinbreastcancer,weexaminedC-erbB2geneamplificationbyusingdiferentialpolymerasechainreaction(dPCR)in75casesofbreastcancer.C-erbB2amplificationwasfoundin34.7%(26/75)patientsandwassignificantincas-esofnegativeERorinpresenceofmorethan3metastaticlymphnodes.Norelationshipwasnoticedbe-tweentheotherfactorsandtheamplification.Thesamesamplesweretestedagainbysouthernblothy-bridizationtoascertainC-erbB2amplificationandtheresultswerenearlythesameasabove,includingonecaseofrearrangement.C-erbB2amplificationtestedbydifferentialPCRisquick,simple,reliableandhasnoradioactivespot,whichishelpfulforpredicatingprognosisandmetastaticpotentialoftu-mour.Itisvaluabletousethemethodinclinicalpractice.
