摘要
以酶组织化学、分子杂交及放免法观察了慢性阻塞性肺疾病(COPD)患者和对照者肺组织中一氧化氮合成酶(NOS)的分布、mRNA表达及活性。结果表明:NOS广泛分布于对照组支气管、肺泡管和部分肺泡囊上皮细胞及肺血管内皮;COPD组支气管上皮细胞及肺小动脉内皮NOS活性较对照组低,但在肺间质和肺血管周围出现NOS阳性细胞增多;斑点和Northern印迹杂交显示COPD组肺组织NOS基因mRNA水平较对照组明显低;COPD组肺组织和肺小动脉环磷酸鸟苷含量均较对照组明显降低(P<0.01)。提示一氧化氮在肺脏中发挥重要生理作用及在COPD发病中可能具有重要意义。
sing NADPH-diaphorase histochemical method,Northern blot analysis,and radioim-munoassay,we have studied the distribution and activity as well as mRNA expression of nitric ox-ide synthase(NOS)in lungs of 8 patients with chronic obstructive pulmonary disease(COPD)and 6 without COPD. The data showed that 1.NOS is present in epithelial cells of bronchi,alveo-lar duc,and alveolar sac;and in endothelium,but rare in media and adventitia of pulmonary ves-sels in patients with or without COPD. However,the activity of NOS markedly decreased in pa-tients with COPD as compared with that in those without COPD,2.The mRNA expression ofNOS in patients with COPD(0.34±0.11U)was significantly lower than that in those withoutCOPD(0.84 ± 0.19U)(P<0.01)3.cGMP level of lung tissues in patients with or withoutCOPD was43.8±5.2 and 86.6±7.5pmol/g wet wt.respectively(P<0.01),and that of pul-monary small arteries in patients with or without COPD was 56.8± 6.4 and 89.6±8. 6pmol/gwet wt.respectively(P<0.01).These results indicate that nitric oxide may play an importantrole in physiology of lung and pathogenesis of COPD.
