摘要
目的建立大孔树脂富集蟾皮总二烯内酯的工艺。方法蟾皮醇提液减压回收醇,经石油醚脱脂,水层上DM130大孔树脂,不同浓度乙醇梯度洗脱,TLC显色结合HPLC法检测二烯内酯所在洗脱部位,比色法测定总二烯内酯含量。结果蟾皮中二烯内酯集中在70%乙醇洗脱部分;采用本实验确定的工艺,制得蟾皮总二烯内酯含量为53.90%,收率为0.23%。结论该工艺可以较好地富集蟾皮总二烯内酯。
Objective To establish processing method for enriching total bufadienolides from the skin of toad by maeoroporous resin. Methods The alcohol extraction of the skin of toad was separated by water - petroleum ether phases after eliminating alcohol. Then the water part was added to the column of DM130 maeoroporous resin, and eluted gradually by alcohol. The total bufadienolides was identified to be in which part by HPLC and TLC, and the content was determined by UV, teloeinobufagin as reference substance, at 299nm. Results The total bufadienolides was gathered in 70% aleohcd. The content of total bufadienolides was 53.90%, the extracting rate was 0.23%. Conclusion The processing technology can he used to enrich total bufadienolides from the skin of toad.
