摘要
目的:研究外源性人甲状旁腺素(1-34)hPTH(1-34)对原代培养新生Wistar鼠心肌成纤维细胞(CFs)增殖及胶原合成的影响。方法:分离、培养Wistar乳鼠心肌成纤维细胞,加入不同浓度的hPTH(1-34)共培养,四氮唑盐比色法(MTT)、3H-TdR掺入法检测细胞增殖;3H-Proline掺入法测定胶原合成。结果:随着一定浓度hPTH(1-34)的升高,CFs MTT法A490值及3H-TdR的掺入量、3H-脯氨酸掺入率呈明显的递增趋势(P<0.01);维拉帕米预处理组与单独应用组10-8mol/L hPTH相比,以上指标均显著降低(P<0.01)。结论:PTH可显著刺激成纤维细胞增殖及胶原合成,L型钙通道开放引起的细胞外钙内流增加为其重要机制之一。
Objective:To investigate the effects of parathyroid hormone (PTH) on proliferation and collagen synthesis of rat cardiac fibroblasts (CFs), Methods:Isolated and cultured CFs of Wistar rats were cultured with PTH. CFs proliferation was measured by MTT assay, The DNA synthesis of CFs was measured by 3 H - TdR uptake test. Collagen synthesis was measured by 3 H - Proline incorporation. Results:A490 value of CFs by MTT assay , CFs proliferation and the synthesis of collagen increased graduately with the elevation of hPTH concentrations was enhanced remarkably by constantly action of different concentration of hPTH(1-34) incubation(P〈 0.01); Compared to the group in which 10^-8mol/L hPTH was used separatdy, group pretreated with 10^-5mol/L verepamilshowed a significant decrease in index above(P〈 0.01 ). Conclusion: PTH dose - dependently increased DNA and collagen synthesis of CFs, partly due to augmented entry of calcium into call via voltage - dependent Ca^2 + channel.
