摘要
目的:探讨复方鳖甲软肝片对慢性肾衰竭(CRF)大鼠肾组织转化生长因子β1(TGF-β1)表达的影响。方法:采用5/6肾切除法制作大鼠CRF实验动物模型。将大鼠随机分为7组:正常组、假手术组、模型组、蒙诺组和复方鳖甲软肝片低、中、高剂量组。术后2个月处死大鼠,用肾小球硬化指数(GSI)和肾小管-间质损伤评分来评价各组大鼠肾组织病理改变,并应用免疫组化法检测肾组织TGF-β1的表达。结果:模型组与正常组和假手术组相比,GSI和肾小管-间质损伤评分明显升高(P<0.05),肾组织TGF-β1表达显著增加(P<0.05);复方鳖甲软肝片低、中、高剂量组和蒙诺组与模型组相比,GSI和肾小管-间质损伤评分明显降低(P<0.05),肾组织TGF-β1表达明显减少P<0.05);复方鳖甲软肝片低、中、高剂量组、蒙诺组四组间在GSI、肾小管-间质损伤评分和TGF-β1表达方面比较均无统计学差异(P>0.05)。结论:复方鳖甲软肝片能下调CRF大鼠肾组织TGF-β1的过度表达,减轻肾脏病理组织学损伤,这一下调作用可能是该药治疗CRF和抗肾纤维化的机制之一。
Objective: To investigate the effect of complex Biejia Ruangan tablet (CBRT) on renal transforming growth factor-β1 (TGF-β1) expression in rats model with chronic renal failure(CRF). Methods: 5/6 nephrectomized rats animal model was used. The rats were randomly divided into 7 groups:normal group, sham operation group, model group, Fosinopril treatment group, CBRT low, middle and high dose treatment groups. The rats were killed two months after operation. Renal tissue were examined by light microscopy and evaluated by glomemloselerosis index(GSI) and the score of renal tubular - interstitium injury. Immunohistochemistry was applied to measure the renal expression of TGF-β1. Results: In comparision with normal group and sham operation group, GSI, the score of renal tubular- interstitium injury and the renal expression of TGF-β1 of model group all markedly increased ( P 〈 0.05). In eomparision with model group, GSI, the score of renal tubularinterstitium injury and the renal expreKsion of TGF-β1 of CBRT low, middle and high dose treatment groups and Fosinopril treatment group all markedly decreased (P〈 0.05), no significant difference was detected between four treatment groups (P 〉0.05). Conclusion: CBRT can decreased the renal expression of TGF-β1 in rats with CRF and the renal tissue injury. And maybe CBRT improves CRF and suppresses renal fibrosis development in part via down - regulating TGF-β1 expression.
