摘要
利用激光扫描共聚焦显微镜的高功率激光对标记CFDA荧光探针的正常大鼠体外培养心肌细胞进行照射。通过荧光漂白后荧光重新分布技术观察大鼠心肌细胞缝隙连接的细胞通讯功能及银杏内酯B对细胞通讯的影响。结果发现体外培养的正常大鼠心肌细胞间存在着缝隙连接,具有细胞间通讯的功能。银杏内酯B对正常大鼠心肌细胞缝隙连接介导的细胞间通讯具有抑制作用。
In present study, we observed effect of ginkgolide B(GB) on functions of gap junctions in rat cardiomyocytes using fluorescence redistribution after photobleaching under laser scanning confocal microscopy. The fluorescence of cells dyeing CFDA was bleached with powerful laser and fluorescence recovery rate was recorded. Results showed that fluorescence recovery in rat cardiomyocyte appeared after bleaching. GB could inhibit fluorescence recovery rate of the cells after laser bleaching. Our data indecated that GB might inhibit cell-cell communication via pathway of changing function of Gap junctions.
