摘要
用化学成分预实验和高效液相色谱法对中药皂角刺中所含的类成分进行了研究。经化学成分预实验得知,皂角刺中含有黄酮、内酯、香豆素或其甙类、甾醇、三萜、酚类、有机酸、还原糖、三萜皂甙等成分。在此基础上,建立了分析皂角刺的高效液相色谱法。即以V(乙腈)∶V(甲酸)=99.5∶0.5的溶液A和V(水)∶V(甲酸)=99.5∶0.5的溶液B作为流动相,检测波长为278 nm,柱温为25℃,进样量为20μL,流速为1 mL/m in,梯度条件为5%A保持5 m in,然后经过58 m in线性升至34%A,然后经过5 m in线性升至100%A,再在100%A保持5m in。确定了提取黄酮类有效成分的较佳工艺为:V(乙醇)∶V(水)=70∶30,加热回流1 h。该文为深入分析皂角刺的化学组成、制备单体化合物及活性实验等提供了参考。
The chemical constituents of Spina Gleditsiae include fiavonoids, lactones, coumarins, hydroxybenzenes,organic acids, saponins, etc. HPLC method was firstly established on the basis of preliminary study. The mobile phases are solvent A of V( acetonitrile):V( formic acid) = 99.5:0.5 and solvent B of V(water) : V( formic acid) = 99.5:0. 5. The mobile phase was adopted by linear gradient elution starting from 5% A for 5 min,5% A to 34% A in 58 rain,34% A to 100% A in 5 min,and 100% A for 5 min, injection volume: 20 μL, flow rate: 1 mL/min, column temperature: 25 ℃ , and detection wavelength:278 nm. The optimized condition for extraction of the effective components is refluxing by V(ethanol) :V(water) =70:30 for 1 h. This gives a good foundation for future works.
