摘要
长效载药微粒可使药物在几周或几个月内以一定速率释放,以维持有效血药浓度,减少给药次数,提高疗效。延长药物在体内的时间和有效控制药物的释放速率是长效载药微粒需要解决的两个基本问题。该文就延长载药微粒在体内的循环时间、延长载药微粒在吸收部位的停留时间、控制药物在释放初期的“突释”3个方面的研究进展进行了综述,并对长效微粒释药系统研究中的相关问题进行了讨论。引用文献42篇。
Long-lasting microparticles have proven to be very useful for sustained and controlled drug delivery. They can achieve drug release over sustained periods of weeks or months, to maintain the drug blood level in a desired therapeutic range. Two basic issues that should he considered are drug residence time and release rate control. For such drug systems, another serious problem is the initial burst release, which is not desirable in most of the sustained-drug delivery applications. Recent advances of long-lasting microparticles in prolonging the circulating time in vivo and residence time at the absorption site, and the solutions to the initial burst release are discussed in this review. In addition, related problems and development trends of drug delivery are analyzed. 42 refers are cited.
