摘要
作者应用DNA重组技术,将去信号肽的重组人x2a型干扰素(IFN)基因分别克隆入具有不同启动子的表达载体,转化相应的宿主菌后表达,结果显示具有P_RP_L双重启动子的表达载体表达x2a IFN最高,滴度可达22×10~9IU/L菌液。具有trp启动子的表达载体经β-吲哚乙酸诱导后,表达x2a IFN的水平可增加3~5倍,
The human intcrferon (IFN) alpha 2a gene was inserted into 6 different expression vectors to be expressed under control of Tac, Trp, Trc and tandem PRPL promoters respectively, and then transformed into different host cells. The results show that the expression vactor whith a tandem PRPL promoter has a higher expression level of IFN alpha 2a than others. The expression under theregulation of this tandem promoter may reach 2.2×109IU/L,which may be used for large scale production , The different length between the SD sequence and start codon ATG affects the expression to some extent
