摘要
[目的]探讨超氧化物歧化酶(SOD)对染矽尘大鼠肺泡巨噬细胞MMP-2、MMP-9表达的影响。[方法]对雄性Wistar大鼠行支气管肺泡原位灌洗术,获取肺泡巨噬细胞。将贴壁后的肺泡巨噬细胞分为对照组、模型组和干预组,均用无血清DMEM培养基孵育。模型组加入SiO2粉尘悬液,干预组用SOD预处理1h后再加入SiO2粉尘悬液。各组肺泡巨噬细胞分别孵育1、2、4、8、16h,用免疫细胞化学和RT-PCR方法观察其MMP-2、MMP-9蛋白及其mRNA的动态变化。[结果]模型组肺泡巨噬细胞MMP-9蛋白及其mRNA表达在1、2、4、8h均较对照组明显增强(P<0.01),分别是对照组的1.229、1.358、1.401、1.354倍和1.402、1.637、1.705、1.463倍。MMP-2蛋白及其mRNA表达无明显变化。干预组肺泡巨噬细胞MMP-9蛋白及其mRNA表达在1、2、4、8h较模型组显著减弱(P<0.05),但仍高于对照组(P<0.05)。[结论]应用SOD可降低染矽尘大鼠肺泡巨噬细胞MMP-9蛋白及其mRNA的表达。
[ Objective ] To explore the effects of SOD on the expression of MMP-2, MMP-9 in alveolar macrophage of rats exposed to SiO2. [ Methods ] Alveolar macrophages obtained from bronchoalveolar lavage fluent were divided into control group, model group and intervention group. SiO2 was given to stimulate the alveolar macrophage in model group and intervention group, and SOD was added before giving SiO2 in intervention group. The protein and mRNA expression of MMP-2, -9 of alveolar macrophages were evaluated by immunocytochemistry and RT-PCR. [ Results ] ( 1 )Expression of MMP-9 in alveolar macrophages of model group were higher than that of control group in 1, 2, 4, 8 hours. ( 2 ) Expression of MMP-9 in alveolar macrophages of intervention group were lower than that of model group in 1, 2, 4, 8 hours, but still higher than that of control group. [ Conclusions ] SOD could decrease the expression of MMP-9 in alveolar macrophages.
