呋喃二氢吡啶二羧酸酯的抗实验性心律失常作用 被引量:1

摘要 呋喃二氢吡啶二羧酸酯(EFDP)9mg/kg iv可明显对抗乌头碱诱发大鼠的心律失常,转窦率为100%;使CaCl_2所致的大鼠室颤发生率降低86%;结扎大鼠左冠状动脉引起的心律失常发生率降低83%;并能提高豚鼠心脏对哇巴因中毒时的耐量,与溶剂对照组比较,使出现室早、室速、室颤和心脏停搏时的哇巴因用量分别增加50,66,68和59μg。给氯仿-肾上腺素的6只家兔用iv EFDP4.5mg/kg则均未发生心律失常。EFDP 23~46mg/kg ip使氯仿引起的小鼠室颤发生率分别减少53%和73%。EFDP 19.6μmol/L冠脉给药,离体兔心在缺血再灌注期间无心律失常发生。结果表明,EFDP具有显著的抗实验性心律失常作用。 Diethyl 2,6 -dimethyl - 4 (furnyl)- 1,4- dihydropyridine -3,5- dicarboxylate (EFDP ) 9 mg/kg was intravenously injected, significantly counteracted the arrhythmias induced by aconitine.The incidence of recovering sinus rhythm was 100%; the incidence of ventricular fibrillation induced by CaCl2 decreased by 86%; the incidence of arrhythmias induced by ligation of the left coronary artery decreased by 83% in rats; and also increased the tolerant dose of ouabain toxkation in guinea pigs. Compared with the solvent control group, doses of ouabain to produce ventricular premature beats, ventricular tachycardia, ventricular fibrillation and cardiac arrestin creased by 50, 66, 68 and 59 μg, respectively. EFDP 4.5 mg/kg was injected into 6 rabbits,that had been gaven chloroform-adrenaline intravenously did not cause arrhythmias. Ip EFDP 23-46 mg/kg, the incidence of ventricular fibrillation induced by chloroform reduced by 53% and 73% in mice respectively. After EFDP 19.6 μmol/L was infused into the coronary artery of isolated rabbit hearts arrhythmias did not occur during ischemia and reperfusion. The results indicate that EFDP may significantly prevent experimental arrhythmias.
