活性氧对ECV304细胞生长和凋亡的相关基因表达谱研究 被引量:3

摘要 目的:探讨活性氧(ROS)对ECV304细胞生长和凋亡作用的相关基因表达谱。方法:采用MTT法分别观察不同浓度AngⅡ在4 h、12 h和24 h时对ECV304细胞生长率和ROS生成量的影响。应用基因芯片技术和光镜分别检测0.0625和1μmol/L AngⅡ作用12 h时ECV304细胞生长作用的相关基因表达谱和细胞形态学变化。结果:一定浓度的AngⅡ作用12 h时ECV304细胞生长率明显增加(P<0.05);基因芯片技术分析显示,在0.0625μmol/L AngⅡ作用ECV304细胞12 h时,与细胞生长相关的ERK、CDK、CCN、PCNA等25个基因普遍上调,而与细胞凋亡相关的DR6、caspase-6、BAK1、PDCD8等12个基因普遍下调,在1μmol/L AngⅡ作用12 h时,呈相反结果。结论:AngⅡ是ECV304细胞产生ROS(.OH)的诱导剂,可引起多种与ECV304细胞生长和凋亡的靶基因表达;N-乙酰半胱氨酸(N-acetyl-L-cyste ine,NAC)可以明显抑制AngⅡ对ECV304细胞的生长作用;ROS(.OH)可能是AngⅡ诱导ECV304细胞生长和凋亡的主要信号转导分子和基因表达调节分子。
