摘要
目的探讨外周血单个核细胞(PBMCs)干扰素诱导的蛋白10(IP-10)mRNA 表达与系统性红斑狼疮(SLE)活动的关系。方法用逆转录-聚合酶链反应(RT-PCR)检测46例 SLE 患者、20例类风湿关节炎(RA)患者、11例非 SLE 肾损伤患者及20名正常健康人 PBMCs IP-10 mRNA 表达。结果 SLE 活动组 PBMCs IP-10 mRNA 表达水平与非活动组间的差异有统计学意义(P<0.01),活动性狼疮肾炎(LN)组与无肾损伤组之间 PBMCs IP-10 mRNA 表达水平的差异亦有统计学意义(P<0.05),而 RA 组和非 SLE 肾病组患者与正常对照组水平相近似(P>0.05)。血清 IP-10水平与PBMCs IP-10 mRNA 表达高度正相关(r=0.8971,P<0.01)。PBMCs IP-10 mRNA 表达水平与狼疮病活动指数(SLEDAI)评分呈正相关(r=0.683 7,P<0.01),与肾 SLEDAI 评分亦高度正相关(r=0.726 0,P<0.01)。结论 SLE 活动时期 PBMCs IP-10 mRNA 表达增加,与 SLE 疾病活动状况相关,尤其与 LN 的活动状况密切相关。
Objective To explore the relation between the expression of PBMCs IP-10 mRNA and systemic lupus erythematosus. Methods The expression of PBMCs IP-10 mRNA was investigated by RT-PCR semi quantitative method and samples from 46 patients with SLE, 20 patients with RA, 11 non-SLE patients with renal impairment and 20 healthy volunteers. Results The expression of PBMCs IP-10 mRNA in active SLE group was significantly higher than that in inactive group( P 〈 0.01 ), and there was significant difference between the active LN group and the no-renal involvement group ( P 〈 0. 05 ). Low expression levels of PBMCs IP-10mRNA were showed in diseased controls and the healthy control ( P 〉 0. 05 ). Serum levels of IP-10 were highly correlated with the expression levels of PBMCs IP-10 mRNA(r =0. 897 1, P 〈 0. 01 ). Furthermore, there was a close correlation between the expression of PBMCs IP-10mRNA and the overall SLEDAI score ( r =0.683 7, P 〈 0.0 1 ) and the SLEDAI renal score ( r =0. 726 0, P 〈0. 01 ). Condusion The expression of PBMCs IP-10 mRNA is significantly correlated with the activity of SLE, and closely correlated with the activity of LN, especially.
